So I took the second strategy for tracking the MT end. The principle of the MT edge detection is: calculate the slope of a certain range of Intensity profile, and select a region where the slope is closest to the one sampled in the first frame. I tried several different algorithms and the best was to list the value in acending order, and average 4 to 5 minuimum positions to determin the ege. Developmet took one week.
